    On the Rapoport-Zink space for GU(2,4)\mathrm{GU}(2, 4) over a ramified prime

    In this work, we study the supersingular locus of the Shimura variety associated to the unitary group GU(2,4)\mathrm{GU}(2,4) over a ramified prime. We show that the associated Rapoport-Zink space is flat, and we give an explicit description of the irreducible components of the reduction modulo pp of the basic locus. In particular, we show that these are universally homeomorphic to either a generalized Deligne-Lusztig variety for a symplectic group or to the closure of a vector bundle over a classical Deligne-Lusztig variety for an orthogonal group. Our results are confirmed in the group-theoretical setting by the reduction method \`a la Deligne and Lusztig and the study of the admissible set

    Didattica ibrida e insegnamento universitario: linee guida per una progettazione efficace

    The paper aims to highlight the role of network and mobile technologies in enhancing the particular characteristics of hybrid instruction solution (HIS) with a view to (a) potentiating/enriching the teaching/learning processes in higher education, (b) exploiting the varied opportunities it offers for their observability, and hence for their monitoring addressed to formative and summative assessment. A possible breakdown of HIS into its key dimensionsof the learning process and the learning space will be proposed. The aim is tounderstand how the characteristics of each dimension can be used to enhance both the teaching/learning and the assessment processes. In this sense, the article will show how this potential can only be captured by solidly integrating the process of instructional design with that of monitoring and assessment.To conclude, the discussion will focus on the different role and responsibility of teachers in a hybrid instruction solution, shifting from a vertical model of knowledge transmission to a more horizontal one, based on collaborative processes as well as individual study.Scopo dell’articolo è mettere in evidenza il ruolo delle tecnologie mobili e di rete nell’esaltare le peculiarità delle hybrid instruction solution (HIS) dal punto di vista (a) del potenziamento/arricchimento dei processi di insegnamento/apprendimento, (b) delle molteplici angolature che offre alla loro osservabilità e quindi al loro monitoraggio e valutazione (formativa e sommativa). Verrà proposta una possibile articolazione delle HIS lungo le dimensioni del processo di apprendimento e degli spazi in cui questo si sviluppa, con l’obiettivo di comprendere come far leva sulle loro rispettive peculiaritàai fini didattici e valutativi. In tal senso, si illustrerà come tali potenzialità possano essere colte solo attraverso una forte integrazione del processo di progettazione didattica con quello di monitoraggio e valutazione. A conclusione dell’articolo si farà riferimento al cambiamento del ruolo del docente nel passaggio da un insegnamento d’aula a un insegnamento di tipo ibrido (h-teaching)

    p66Shc deficiency in the EÎĽ-TCL1 mouse model of chronic lymphocytic leukemia enhances leukemogenesis by altering the chemokine receptor landscape

    The Shc family adaptor p66Shc acts as a negative regulator of proliferative and survival signals triggered by the B Cell Receptor and, by enhancing the production of reactive oxygen species, promotes oxidative stress-dependent apoptosis. Additionally, p66Shc controls the expression and function of chemokine receptors that regulate lymphocyte traffic. Chronic lymphocytic leukemia cells have a p66Shc expression defect which contributes to their extended survival and correlates with poor prognosis. We have analyzed the impact of p66Shc ablation on disease severity and progression in the mouse model of chronic lymphocytic leukemia EÎĽ-TCL1. We show that EÎĽ-TCL1/p66Shc-/- mice develop an aggressive disease that has an earlier onset, a higher incidence and leads to earlier death compared to EÎĽ-TCL1 mice. EÎĽ-TCL1/p66Shc-/- mice display substantial leukemic cell accumulation in both nodal and extranodal sites. The target organ selectivity correlates with an upregulation of chemokine receptors whose ligands are expressed therein. This also applies to chronic lymphocytic leukemia cells, where chemokine receptor expression and extent of organ infiltration were found to inversely correlate with their p66Shc expression levels. p66Shc expression declined with disease progression in EÎĽ-TCL1 mice and could be restored by treatment with the Bruton tyrosine kinase inhibitor Ibrutinib. Our results highlight p66Shc deficiency as an important factor in chronic lymphocytic leukemia progression and severity and underscore p66Shc expression as a relevant therapeutic target

    Efficacy of bendamustine and rituximab as first salvage treatment in chronic lymphocytic leukemia and indirect comparison with ibrutinib: A GIMEMA, ERIC and UK CLL FORUM study

    We performed an observational study on the efficacy of bendamustine and rituximab (BR) as first salvage regimen in chronic lymphocytic leukemia (CLL). In an intention-to-treat analysis including 237 patients, the median progression-free survival (PFS) was 25 months. The presence of del (17p), unmutated IGHV and advanced stage were associated with a shorter PFS at multivariate analysis. The median time-to-next treatment was 31.3 months. Front-line treatment with a chemoimmunotherapy regimen was the only predictive factor for a shorter time to next treatment at multivariate analysis. The median overall survival (OS) was 74.5 months. Advanced disease stage (i.e. Rai stage III-IV or Binet stage C) and resistant disease were the only parameters significantly associated with a shorter OS. Grade 3-5 infections were recorded in 6.3% of patients. A matched-adjusted indirect comparison with ibrutinib given second-line within Named Patient Programs in the United Kingdom and in Italy was carried out with OS as objective end point. When restricting the analysis to patients with intact 17p who had received chemoimmunotherapy in first line, there was no difference in OS between patients treated with ibrutinib (63% alive at 36 months) and patients treated with BR (74.4% alive at 36 months). BR is an efficacious first salvage regimen in CLL in a real-life population, including the elderly and unfit patients. BR and ibrutinib may be equally effective in terms of OS when used as first salvage treatment in patients without 17p deletion. (Registered at clinicaltrials.gov identifier: 02491398)
